Micropropagation of wild cherry (Prunus avium L.) in vitro

Prunus avium is very important part of interspecific diversity of European forests and is high-valued wood. This genetic resource is endangered due to uncontrolled felling and noncontroled hybridization with sweet cherry. An attempt was made to develop method culture in vitro thus to contribute conservation of this genetic resource.
Induction of apical and axillary buds of mature wild cherry tree carried out on the MS medium, with 0,5 mg/1 BAP and 0,lmg/1 IBA growth regulators added, was very successful and almost in 80% of buds were opened and regenerated into shoots. Multiplication of shoots on the same medium was almost tripled after third cycle.
A number of leaves from these plants were regenerated on DCR medium containing 0,5 mg/1 BAP and 0,1 mg/1 NAA or 0,5 mg/1 TDZ and 0,1 mg/1 NAA. Better results were achieved in series with BAP (regeneration was successful in 73%).
Rooting on basal MS medium, supplemented with 126 mg/1 PG and 3 mg/1 IBA, was successful only 20%. Apparently further research is needed to increase knowledge of rooting process.
