Micropropagation and somatic embryogenesis of black locust (Robinia pseudoacacia L.)

Abstract

Initial explants (axillary and apically buds meristems) of black locust (Robinia pseudoacacia L.) were cultured in agar MS medium supplemented with BA (0,5 mg/1), adenin sulfate (20 mg/1) and NAA (0,02 mg/1). Response of initial explants in this medium was good. High rate of multiplication of nodal segments was obtained in the same medium. The root system was regenerated in 100% in agar MS medium supplemented with 0,5 mg/1 IBA. Rooted plants were transferred, after period of acclimatisation, to mixture of humus, sand, perlite and quartz sand (5:3:1:1).

Results of the experiment show that is possible to obtain technological procedure for mass multiplication of black locust by tissue culture technics.

Somatic embryogenesis of black locust were obtained from non-embryogenic explants. Somatic embryos were differentiated indirectly from embryogenic cell suspension callus initiated from shoots. Callus cell suspension were cultured on the liquid MS basal medium supplemented with 8 mg/1 2,4D plus 0,5 mg/1 BA at the beginning of the experiment, and 0,3 mg/1 2,4 D plus 1 mg/1 BA in the later phases. Developing embryogenic structure in this medium were transferred on media lacking hormones. Removal of 2,4D from the culture medium seems to be very efficient. If initial explants are properly selected as well as combination and concentration of growth regulators somatic embryogenesis can be quickly obtained.
